<b>The Mechanism and Regulation of Motor Proteins</b><br /><br />

The intracellular transport is specialized to an extraordinary degree, enabling the delivery of critical cargos to many destinations far from the cell center. This process is driven by kinesin and dynein motors and regulated by cargo adaptors and microtubule-associated proteins (MAPs). We still do not know how cargos decide which direction to move along the microtubule, and how hundreds of different cargos are sorted and delivered to different destinations inside cells. My laboratory uses biochemical reconstitution, structural biology, and singlemolecule imaging methods to understand the mechanism of bidirectional transport of cargos along the microtubules. I will present a biophysical model for the activation, assembly, directionality, and force generation of functional dynein motors. I will also discuss our recent efforts to dissect how opposing motors are coordinated when they are recruited to their respective cargos during transport.<br /><br />
