ENGINEERING FRET-BASED LABELS FOR MULTIPLEXED SINGLE-MOLECULE SENSING This dissertation introduces FRETfluors, a novel class of DNA-based fluorescence tags that enhance multiplexed single-molecule detection using Förster Resonance Energy Transfer (FRET). Integrated with the Anti-Brownian Electrokinetic (ABEL) trap, this approach enables the simultaneous identification of up to 30 molecular species at sub-picomolar concentrations, surpassing current fluorescence multiplexing limits. Beyond fundamental advancements, FRETfluors improve sensitivity and multiplexing in biomedical assays and imaging, streamlining biomarker detection and molecular profiling. These innovations position FRETfluors as a powerful tool for next-generation diagnostics and super-resolution imaging.
